(Nasdaq: PRME), a biotechnology company committed to delivering a new class of differentiated one-time curative genetic therapies, today announced the publication of Phase 1/2 clinical data with PM359, the Company’s investigational autologous hematopoietic stem cell product for p47phox chronic granulomatous disease (CGD) in the New England Journal of Medicine (NEJM). The data will also be presented in a poster session at the 67th American Society of Hematology (ASH) Annual Meeting, December 6-9, 2025 in Orlando, Florida.

The publication, titled “Prime Editing for p47-phox Chronic Granulomatous Disease,” reports initial data for two patients treated in the Phase 1/2 trial of PM359, which was designed to assess safety, biological activity and preliminary efficacy in adult and pediatric study participants. Both patients experienced rapid neutrophil and platelet engraftment, as well as durable restoration of NADPH oxidase activity and early clinical benefit, without any safety concerns. Together, these results provide the first-in-human demonstration of the safety and efficacy of Prime Editing, and support the potential for PM359 as a precise therapeutic strategy for CGD:

  • Both patients enrolled in the study had a history of prior CGD-defining complications, including CGD-associated colitis (CAC), and skin and soft tissue infections, and both were maintained on long-term prophylactic therapy.
  • Both patients experienced rapid neutrophil engraftment, achieving 69% and 83% dihydrorhodamine-positive (DHR+) neutrophils by Day 30, respectively, far in excess of the 20% projected minimum threshold for clinical benefit. DHR activity remained stable over time in both patients, suggesting that gene correction occurred in the long-term repopulating hematopoietic stem cells (HSCs) of the bone marrow.
  • Both patients remain free of new CGD-related complications or significant intercurrent illnesses post-infusion; additionally, Patient 1 stopped his mesalamine treatment and has not experienced a flare of CAC, and Patient 2’s levels of fecal calprotectin have decreased substantially, and his chronic CAC symptoms have abated.
  • No clinically significant adverse events attributable to PM359 occurred in either patient, and all observed toxicities were consistent with busulfan-based conditioning.

Longer-term analyses reinforce duvelisib’s activity across treatment-experienced PTCL populations, including patients with ≥3 prior lines of therapy, with no new safety signals emerging over time. 

Findings further strengthen rationale for ongoing global Phase 3 TERZO study in nodal TFH lymphoma.

BERKELY HEIGHTS, N. J., Dec. 07, 2025 (GLOBE NEWSWIRE) — Secura Bio, Inc. (www.securabio.com), an integrated pharmaceutical company maximizing commercial outcomes for oncology medicines, today presented extended follow up analyses examining additional safety data and impact of prior lines of therapy from the Company’s Phase 2 PRIMO trial (NCT03372057) at the 2025 American Society of Hematology (ASH) Annual Meeting. The results continue to show that duvelisib delivers meaningful clinical activity with a manageable safety profile, even in heavily pretreated patients, underscoring its potential role across a broad PTCL population.

The poster, titled “Duvelisib in patients with relapsed/refractory peripheral T-cell lymphoma: from the Phase 2 PRIMO trial – impact of prior therapy and expanded safety analysis”, was presented by Professor Pier Luigi Zinzani, Head of Hematology at the University of Bologna/Italy, and highlighted new analyses of the dose expansion phase of the PRIMO trial, which included a group of 123 patients who had received at least 2 cycles of one standard regimen. The PRIMO clinical trial was a global, multicenter, open-label, single-arm trial that evaluated the investigational use of duvelisib for the treatment of adult patients with R/R PTCL.

“PTCL patients often cycle through multiple treatments with little improvement in outcomes,” said Prof. Zinzani. “Seeing consistent efficacy even in those with ≥3 prior lines, and without emergent safety concerns at the recommended 75 mg lead-in dose, is highly encouraging. These results reinforce duvelisib’s therapeutic potential across this difficult-to-treat population.”

In the PRIMO expansion phase duvelisib was dosed at 75 mg twice daily for two cycles and followed by 25 mg twice daily, and the primary endpoint was Overall Response Rate (ORR).

New extended analyses demonstrated there was no consistent impact on efficacy outcomes for patients related to number of prior lines of treatment. In patients with 1, 2, and ≥3 prior lines of therapy, outcomes were as follows, respectively: ORR was 29%, 66%, 49%; complete response (CR) was 18%, 52%, 32%; median duration of response (DOR) was 6.5, 11.7, 7.9 months; median progression-free survival (mPFS) was 1.9, 9.0, 3.0 months, and median overall survival (OS) was 30.2, 22.7, 7.3 months. Interestingly, patients with more heavily pretreated disease demonstrated numerically favorable outcomes.

In the analyses, 68% percent of patients received prior CHOP-based therapy, 35% had salvage chemotherapy, 20% had autologous stem cell transplant, 17% had an HDAC inhibitor, and 38% had prior exposure to brentuximab vedotin.

Additionally, expanded safety analyses based on the number of dose cycles, where 123 patients received ≤2 cycles, 63 patients received >2 to 6 cycles, and 25 patients continued on to receive >6 cycles, revealed there was no consistent pattern of higher rates of persisting or emerging AEs with longer treatment duration. Rates of diarrhea showed a modest trend of increase, but there was no increase in rates of colitis.

Adverse events (AEs) in ≥20% of patients by treatment duration included: neutropenia, aminotransferase increase, diarrhea, thrombocytopenia, leukopenia, and fatigue. Grade ≥3 adverse events in ≥5% of patients included: neutropenia, aminotransferase increase, maculopapular rash, thrombocytopenia, lymphopenia, diarrhea, hypokalemia and hypoxia.

“These new analyses capture important insights on the many different profiles and treatment experiences of PTCL patients in our trial and demonstrate the efficacy and safety of duvelisib, regardless of baseline treatment characteristics and prior lines of treatment across the patient population,” said Dr. Christiane Langer, SVP, Head of Clinical and Medical Affairs at Secura Bio. “Secura Bio is committed to expanding the potential of this therapy and other existing oncology medicines to new cancer indications so that more patients have an opportunity to explore a medicine that may truly benefit them and extend their life.”

Previously, final data presented last year at ASH show outcomes including an ORR of 48% (59/123), with a CR rate of 33.3% (41/123). The mDOR was 7.9 months, the mPFS was 3.4 months with a mPFS by baseline histology of 8.3 months (AITL), 3.4 months (PTCL-NOS), and 1.6 months (ALCL), and the mOS was 12.4 months. The most common baseline histologies were PTCL-NOS, AITL, and ALCL. ORRs by baseline histology were 62.2% (AITL), 49.1% (PTCL-NOS), and 15.0% (ALCL). Efficacy in the AITL group was notably favorable, with AITL subgroup outcomes as follows: ORR (62%), CRR (51%), mDOR 11.7 months, mPFS 8.3 months, and mOS 18.1 months. The strength of these findings directly informed the ongoing Phase 3 TERZO trial (NCT06522737) in relapsed/refractory nodal T-follicular helper cell lymphoma (nTFHL), now actively enrolling at over 40 centers in Europe.

About Peripheral T-cell Lymphoma
Peripheral T-cell lymphoma (PTCL) is a rare, aggressive type of non-Hodgkin lymphoma that develops in mature white blood cells that circulate through the bloodstream and lymphatic system. PTCL accounts for between 10-15% of all non-Hodgkin lymphomas and generally affects people aged 60 years and older. Although there are many different subtypes of PTCL, they often present in a similar way, with widespread, enlarged, typically painless lymph nodes in the neck, armpit, and/or groin. There are currently no well-established standards of care for patients with relapsed or refractory disease.

About nodal T-Follicular Helper Cell Lymphoma

Nodal T-follicular helper cell lymphomas (nTFHL) comprise a group of aggressive non-Hodgkin lymphomas that share common genetic, clinical, and cell of origin features. This group of lymphomas originates from a type of white blood cell called a T-follicular helper cell. nTFHLs generally occur in people aged 60 and older and are slightly more common in males. nTFHLs most commonly present with widespread, enlarged, typically painless lymph nodes in the neck, armpit, and/or groin, in addition to rashes, B-symptoms, and immune dysregulation. There are currently no well-established standards of care for patients with relapsed or refractory disease.

SOUTH SAN FRANCISCO, Calif., Dec. 07, 2025 (GLOBE NEWSWIRE) — ALX Oncology Holdings Inc., (“ALX Oncology” or the “Company”) (Nasdaq: ALXO), a clinical-stage biotechnology company advancing a pipeline of novel therapies designed to treat cancer and extend patients’ lives, today announced that positive data from a Phase 2 investigator-sponsored trial (IST) of evorpacept, in combination with standard-of-care rituximab and lenalidomide, for patients with indolent B-cell non-Hodgkin lymphoma (iNHL) is being presented Sunday, December 7 during a poster presentation at the American Society of Hematology (ASH) Annual Meeting 2025 in Orlando, Florida.

The clinical trial conducted by Dr. Paolo Strati, the trial’s lead investigator and Associate Professor of Lymphoma-Myeloma at The University of Texas MD Anderson Cancer Center, along with his colleagues enrolled a total of 24 patients with previously untreated iNHL, 14 patients with follicular lymphoma and 10 patients with marginal zone lymphoma. The primary objective of best CR rate above 80% was met with evorpacept added to R2 in the context of a historical R2 CR rate of 50%. The investigators found the addition of evorpacept to R2 to be a well-tolerated frontline non-chemotherapy regimen for patients with iNHL, resulting in a high CR rate. 92% of patients achieved a complete response, and 8% achieved a partial response (PR), with the overall response rate (ORR) being 100%. One year progression free survival (PFS) rate was 91%, and one year overall survival (OS) rate was 100%.

Genmab A/S (Nasdaq: GMAB) today announced primary data from the pivotal Phase 3 EPCORE® FL-1 study evaluating fixed duration EPKINLY® (epcoritamab-bysp) in combination with rituximab and lenalidomide (EPKINLY + R2) in adult patients with relapsed or refractory (R/R) follicular lymphoma (FL). The study showed that treatment with EPKINLY + R2 reduced the risk of disease progression or death by 79% (HR 0.21, 95% CI: 0.14-0.31, p<0.0001) compared to standard of care R2. Additionally, the overall response rate (ORR) in patients treated with EPKINLY + R2 was 95% (95% CI: 91.5, 97.4) compared to 79% in patients treated with R2 (95% CI: 73.6, 84.1; P<.0001). The EPCORE FL-1 study results were presented during an oral presentation (abstract 466) at the 67th Annual Meeting and Exposition of the American Society of Hematology (ASH) in Orlando, Florida, featured in the “Emerging Therapies and Immunotherapies in Blood Cancers” ASH press briefing, and have been simultaneously published in The Lancet.

“Patients with relapsed or refractory follicular lymphoma have historically had limited treatment options,” said Lorenzo Falchi, M.D., Lymphoma Specialist, Department of Medicine, Memorial Sloan Kettering Cancer Center. “The EPCORE FL-1 results demonstrate that epcoritamab plus R2 is the first bispecific antibody-based, chemotherapy-free combination to show superior clinical benefit over standard of care in a Phase 3 trial, underscoring its potential to redefine the second-line treatment landscape for follicular lymphoma.”

The EPCORE FL-1 study included patients with R/R FL following at least one prior line of treatment across a broad range of patient characteristics and disease risk factors. Among patients who were treated with EPKINLY + R2 at the second planned interim analysis (median follow-up, 14.8 months), 83% achieved a complete response (CR) (n=201/243, 95% CI: 77.4, 87.3) compared to a 50% CR rate among patients treated with R2 (n=122/245, 95% CI: 43.4, 56.2). The 12-month duration of response (DOR) was 89% (95% CI: 83.6, 93.0) versus 49% (95% CI: 38.8, 57.5) for patients treated with EPKINLY + R2 and R2, respectively.

The safety profile of EPKINLY + R2 in the EPCORE FL-1 study was consistent with the known safety profiles of the individual regimens (epcoritamab and R2). Grade 3 or 4 treatment-emergent adverse events (TEAE) were reported in 90.1% of patients treated with EPKINLY + R2 compared to 67.6% of patients treated with R2, the difference being primarily driven by higher rates of Grade 3 or 4 neutropenia (68.7% vs. 42.0%) and infections (33.3% vs. 15.1%). Fatal TEAEs occurred in 1.6% of patients treated with EPKINLY + R2 compared to 3.8% patients treated with R2. TEAEs leading to discontinuation occurred in 18.9% and 12.2% of patients treated with EPKINLY + R2 and R2, respectively. With the three step-up dosing regimen, CRS events were low grade and occurred in 26.3% of patients (21.2% Grade 1, 5.3% Grade 2).

In November 2025, the U.S. Food and Drug Administration (FDA) approved the combination of EPKINLY + R2 for the treatment of patients with relapsed or refractory FL after one or more lines of systemic therapy. EPKINLY is also approved in the U.S. to treat adults with relapsed/refractory FL after two or more prior treatments.

About the EPCORE® FL-1 Trial
EPCORE® FL-1 (NCT05409066) is a Phase 3 open-label interventional trial to evaluate the safety and efficacy of epcoritamab plus rituximab and lenalidomide (R2) versus R2 alone in patients with relapsed/refractory (R/R) follicular lymphoma (FL). Patients were randomized to receive EPKINLY in combination with rituximab and lenalidomide (n=243) or rituximab and lenalidomide alone (n=245). Patients received EPKINLY in 28-day cycles for a total of 12 cycles or until disease progression or unacceptable toxicity, whichever occurred first. Efficacy was established based on the dual primary endpoints of progression free survival (PFS) and overall response rate (ORR) determined by Lugano 2014 criteria as assessed by Independent Review Committee (IRC). Additional efficacy outcome measures include complete response (CR) and duration of response (DOR).

About Follicular Lymphoma (FL)
Follicular lymphoma (FL) is typically an indolent, or slow-growing, form of non-Hodgkin lymphoma (NHL), that arises from B-lymphocytes. Trial Design and Data Cut Overview

PIONEER is a Phase 1b open-label dose-escalation clinical trial evaluating the safety and efficacy of pociredir, an oral once-daily HbF inducer, in adult patients with severe SCD. The 20 mg cohort of the Phase 1b PIONEER trial includes 12 adults with severe SCD. At the November 11, 2025 data cutoff, all 12 patients completed the Week 6 visit and are included in the Week 6 analyses. Six patients (50%) who enrolled earlier in the cohort reached the Week 12 visit at the time of data cutoff and are included in the Week 12 analyses. Week 6 results therefore reflect all 12 patients, while Week 12 results reflect the first 6 patients to complete the full treatment period. All 12 patients are expected to complete the full 12-week treatment period, and Fulcrum plans to report updated results in Q1 2026.

PIONEER Study 20 mg Dose Cohort Initial Efficacy Data

Initial results from the 20 mg dose cohort of the Phase 1b PIONEER trial (n=12) are as follows:

  • Mean absolute HbF increased by 9.9% at 6 weeks of treatment with pociredir (vs. 5.6% at Week 6 and 8.6% at Week 12 in the 12 mg cohort), increasing from a baseline of 7.1% to 16.9%. As of the November 11, 2025 data cutoff, 7 of 12 patients (58%) achieved absolute HbF levels ≥20% at Week 6, and all patients demonstrated a robust HbF increase. HbF levels of 20% are associated with ~90% of patients experiencing zero VOCs per year, based on real-world data presented by Fulcrum at the 20th Annual Sickle Cell & Thalassemia Conference (ASCAT) in October 2025.
  • A clear dose-response was observed, with a >3.75-fold mean induction of HbF at Week 12 among patients who reached the Week 12 visit as of the November 11, 2025 data cutoff (n=6), compared to a 2.4-fold mean induction at Week 12 in the 12 mg cohort. The average baseline for these six patients is 5.0% as compared to 7.1% for the full cohort. Fold induction accounts for differences in baseline HbF levels and enables a normalized comparison of dose-response.
  • The proportion of F-cells (HbF-containing red blood cells) increased from a mean of 31% at baseline to 58% at Week 6 (n=9), indicating early progression toward pan-cellular HbF induction (evenly distributed across red blood cells). F-cells are resistant to sickling and hemolysis because of HbF-mediated inhibition of sickle hemoglobin (HbS) polymerization. Consequently, higher proportions of F-cells are associated with improved red blood cell health.
  • Markers of hemolysis and erythropoiesis improved at Week 6:
    • Indirect bilirubin decreased by 37% (vs. 37% at Week 12 in the 12 mg cohort)
    • Lactate dehydrogenase (LDH) decreased by 37% (vs. 28% at Week 12 in the 12 mg cohort)
    • Red cell distribution width decreased by 22% (vs. 27% at Week 12 in the 12 mg cohort)
    • Reticulocyte counts decreased by 33% (vs. 31% at Week 12 in the 12 mg cohort), indicating healthier bone marrow function
  • Mean hemoglobin increased by 0.8 g/dL at Week 6 (vs. 0.9 g/dL at Week 12 in the 12 mg cohort), increasing from a baseline of 7.3 g/dL to 8.1 g/dL. Combined with reductions in reticulocyte counts, these findings indicate decreased red blood cell destruction and improvements in anemia.
  • A trend of reduced VOC frequency was observed relative to patients’ documented VOC frequency during the 6–12 months prior to enrollment. As of November 11, 2025 data cut off, eight of 12 patients (67%) reported no VOCs during the treatment period.

Pociredir Safety Update

  • As of the November 11, 2025 data cutoff, pociredir has been dosed in 148 adults, including 89 subjects in multiple dose cohorts up to 12 weeks.
    • 103 healthy subjects, including 44 who received pociredir for 10 to 14 days treatment duration
    • 45 SCD patients who received pociredir for up to 12 weeks treatment duration
  • The safety profile observed in the 20 mg dose cohort as of the November 11, 2025 data cutoff, together with follow-up data from the 12 mg dose cohort, remained consistent with previously reported safety data. About Sickle Cell Disease
SCD is a genetic disorder of the red blood cells caused by a mutation in the HBB gene. This gene encodes a protein that is a key component of hemoglobin, a protein complex whose function is to transport oxygen in the body. The result of the mutation is less efficient oxygen transport and the formation of red blood cells that have a sickle shape. Trial Design and Data Cut Overview

PIONEER is a Phase 1b open-label dose-escalation clinical trial evaluating the safety and efficacy of pociredir, an oral once-daily HbF inducer, in adult patients with severe SCD. The 20 mg cohort of the Phase 1b PIONEER trial includes 12 adults with severe SCD. At the November 11, 2025 data cutoff, all 12 patients completed the Week 6 visit and are included in the Week 6 analyses. Six patients (50%) who enrolled earlier in the cohort reached the Week 12 visit at the time of data cutoff and are included in the Week 12 analyses. Week 6 results therefore reflect all 12 patients, while Week 12 results reflect the first 6 patients to complete the full treatment period. All 12 patients are expected to complete the full 12-week treatment period, and Fulcrum plans to report updated results in Q1 2026.

PIONEER Study 20 mg Dose Cohort Initial Efficacy Data

Initial results from the 20 mg dose cohort of the Phase 1b PIONEER trial (n=12) are as follows:

  • Mean absolute HbF increased by 9.9% at 6 weeks of treatment with pociredir (vs. 5.6% at Week 6 and 8.6% at Week 12 in the 12 mg cohort), increasing from a baseline of 7.1% to 16.9%. As of the November 11, 2025 data cutoff, 7 of 12 patients (58%) achieved absolute HbF levels ≥20% at Week 6, and all patients demonstrated a robust HbF increase. HbF levels of 20% are associated with ~90% of patients experiencing zero VOCs per year, based on real-world data presented by Fulcrum at the 20th Annual Sickle Cell & Thalassemia Conference (ASCAT) in October 2025.
  • A clear dose-response was observed, with a >3.75-fold mean induction of HbF at Week 12 among patients who reached the Week 12 visit as of the November 11, 2025 data cutoff (n=6), compared to a 2.4-fold mean induction at Week 12 in the 12 mg cohort. The average baseline for these six patients is 5.0% as compared to 7.1% for the full cohort. Fold induction accounts for differences in baseline HbF levels and enables a normalized comparison of dose-response.
  • The proportion of F-cells (HbF-containing red blood cells) increased from a mean of 31% at baseline to 58% at Week 6 (n=9), indicating early progression toward pan-cellular HbF induction (evenly distributed across red blood cells). F-cells are resistant to sickling and hemolysis because of HbF-mediated inhibition of sickle hemoglobin (HbS) polymerization. Consequently, higher proportions of F-cells are associated with improved red blood cell health.
  • Markers of hemolysis and erythropoiesis improved at Week 6:
    • Indirect bilirubin decreased by 37% (vs. 37% at Week 12 in the 12 mg cohort)
    • Lactate dehydrogenase (LDH) decreased by 37% (vs. 28% at Week 12 in the 12 mg cohort)
    • Red cell distribution width decreased by 22% (vs. 27% at Week 12 in the 12 mg cohort)
    • Reticulocyte counts decreased by 33% (vs. 31% at Week 12 in the 12 mg cohort), indicating healthier bone marrow function
  • Mean hemoglobin increased by 0.8 g/dL at Week 6 (vs. 0.9 g/dL at Week 12 in the 12 mg cohort), increasing from a baseline of 7.3 g/dL to 8.1 g/dL. Combined with reductions in reticulocyte counts, these findings indicate decreased red blood cell destruction and improvements in anemia.
  • A trend of reduced VOC frequency was observed relative to patients’ documented VOC frequency during the 6–12 months prior to enrollment. As of November 11, 2025 data cut off, eight of 12 patients (67%) reported no VOCs during the treatment period.

Pociredir Safety Update

  • As of the November 11, 2025 data cutoff, pociredir has been dosed in 148 adults, including 89 subjects in multiple dose cohorts up to 12 weeks.

The data is being featured in a poster presentation at the 67th American Society of Hematology (ASH) Annual Meeting and Exposition.

“These updated data from our Phase 1 study continue to highlight a positive safety and efficacy profile of TSC-101 in patients with heme malignancies undergoing allogeneic HCT. All three patients who reached two years of follow-up have no detectable disease as they have remained relapse-free and in complete donor chimerism,” said Chrystal U. Louis, M.D., Chief Medical Officer. “Additionally, there have been no dose-limiting toxicities and patients who received TSC-101 continue to show improved relapse-free and overall survival compared to control-arm patients. Key Presentation Highlights:

Patients in the treatment arm of the ALLOHATM Phase 1 trial (NCT05473910) receive TSC-101 post standard of care HCT, whereas control-arm patients receive standard of care HCT alone. As of the September 19th, 2025 data cut, 42 patients (23 in the TSC-101 treatment arm and 19 in the control arm) were eligible for inclusion in the safety analysis set. The key endpoints in the trial are safety and efficacy, with exploratory endpoints including donor chimerism and minimal residual disease (MRD) status.

  • Relapse-free survival (RFS) (HR=0.50; p=0.23) and overall survival (OS) (HR=0.61; p=0.52) were improved in the treatment arm relative to the control arm.
    • 4 of 19 (21%) treatment-arm patients relapsed compared to 6 of 18 (33%) control-arm patients.
      • One treatment-arm patient with AML experienced disease relapse on day 161 and was given a third dose of TSC-101 without lymphodepletion. The additional administration of TSC-101 resulted in a complete response including complete donor chimerism that was maintained for 5 months.
    • The hazard ratio for probability of relapse was 0.46 (p=0.22).
    • 8 of 37 (22%) patients had TP53 mutations, with 6 cases in the treatment arm and 2 cases in the control arm. Of the 6 patients in the treatment arm, only 1 has relapsed. Both patients with TP53 mutations in the control arm have relapsed and subsequently succumbed to their disease. The first patient with a TP53 mutation to receive TSC-101 has now reached two years of follow-up and remains relapse-free.
  • All 3 (100%) TSC-101-treated patients that reached two years of follow-up remained relapse-free as of the data cutoff, compared to 1 of 4 (25%) patients in the control arm, consistent with effective elimination of residual cancer cells post-HCT and durable remission with TSC-101 infusion.
  • TSC-101 infusions were well-tolerated at all dose levels, with no dose-limiting toxicities. Observed adverse events were similar across the treatment and control arms and were generally consistent with post-HCT adverse events.
  • Mixed chimerism or relapses following TSC-101 infusions were found to be significantly associated with greater ex vivo expansion of TCR-T cells during the manufacturing process. A new commercial-ready process reduces the manufacturing time from 17 days to 12 days and has a significant reduction in ex vivo expansion.

MAINZ, Germany, and ROCKVILLE, USA, December 6, 2025 (GLOBE NEWSWIRE) — BioNTech SE (Nasdaq: BNTX, “BioNTech”) and OncoC4, Inc. (“OncoC4”) today presented data from the non-pivotal dose-confirmation stage of the global randomized Phase 3 trial PRESERVE-003 (NCT05671510) for gotistobart (also known as BNT316 or ONC-392), a tumor microenvironment-selective regulatory T cell (“Treg”) depletion candidate, targeting CTLA-4 in patients with metastatic squamous non-small cell lung cancer (sqNSCLC). Gotistobart demonstrated a clinically meaningful overall survival (“OS”) benefit compared to standard-of-care chemotherapy and a manageable safety profile in sqNSCLC patients whose disease had progressed following anti-PD-(L)1 therapy and platinum-based chemotherapy. Data from the non-pivotal stage of the trial are being presented today in an oral presentation at the IASLC ASCO 2025 North America Conference on Lung Cancer, hosted by the International Association for the Study of Lung Cancer in Chicago, Illinois, USA.

“With a median survival of less than a year, advanced squamous NSCLC remains an aggressive and difficult-to-treat lung cancer1,2. Survival outcomes have improved in recent years due to advances in immunotherapy and combination regimens. However, patients who progress on anti-PD-(L)1 inhibitor treatment face a poor prognosis, leaving them only with the option of chemotherapy or palliative care,” said Byoung Chul Cho, M.D., Ph.D., Lead Investigator and Professor at the Division of Medical Oncology, Yonsei Cancer Center, Seoul. “We are encouraged by the median overall survival still not being reached for patients treated with gotistobart at almost 15 months of follow-up, and we are excited to continue to investigate the candidate’s potential in the ongoing pivotal stage of the trial.”

The analysis from the non-pivotal stage of the global Phase 3 trial included 45 metastatic sqNSCLC patients who received gotistobart as monotherapy, compared with 42 metastatic sqNSCLC patients who received chemotherapy (docetaxel) as second or later line of systemic therapy. At the data cut-off on August 8, 2025, 87 patients with sqNSCLC had been randomized to either gotistobart 6 mg/kg with two 10 mg/kg loading doses (N=45) or docetaxel 75 mg/m2 (N=42). The OS rate at 12 months was 63.1% for gotistobart compared to 30.3% for docetaxel. At a median follow-up of 14.5 months, patients in the gotistobart treatment arm had not yet reached the median OS, while the docetaxel treatment arm achieved a median OS of 10 months. The data showed that the gotistobart arm reduced the risk of death by 54% compared to the docetaxel treatment arm (HR=0.46, 95% CI: 0.25–0.84; nominal p-value 0.0102). The safety profile of gotistobart was consistent with previously established data and remained manageable. Grade ≥3 treatment-related adverse events (“AEs”) were reported in 19/45 (42.2%) patients in the gotistobart treatment arm versus 20/41 (48.8%) patients in docetaxel treatment arm. The pivotal stage of the Phase 3 trial is ongoing in more than 160 sites globally.

About the PRESERVE-003 trial
PRESERVE-003 (NCT05671510) is a two-stage, open-label Phase 3 trial evaluating the efficacy and safety of gotistobart as monotherapy compared to the standard-of-care chemotherapy (docetaxel) in sqNSCLC patients, who have progressed on PD-(L)1 inhibitors and platinum-based chemotherapy. The non-pivotal stage of the trial originally included all NSCLC patients. The ongoing pivotal stage is currently enrolling patients with sqNSCLC. During the ongoing pivotal stage, approximately 500 patients are planned to be enrolled at clinical sites in various countries and regions, including Australia, Belgium, Canada, China, Germany, Italy, the Netherlands, Spain, South Korea, Türkiye, the United Kingdom and the United States. The primary endpoint is overall survival. Secondary endpoints include overall response rate, progression-free survival and safety profile.

About gotistobart (BNT316/ONC-392)
Gotistobart (BNT316/ONC-392) is a tumor microenvironment-selective Treg depletion candidate developed jointly by BioNTech and OncoC4. As a pH-sensitive monoclonal antibody, gotistobart is designed to enable CTLA-4 protein recycling. After binding to the CTLA-4 receptor on the cell surface, the complex is internalized, and the pH change causes the antibody to unbind, allowing CTLA-4 to return to the surface to preserve the immune checkpoint function at peripheral organs and to enhance anti-tumor immunity in the tumor microenvironment3. Non-small cell lung cancer (“NSCLC”) covers all epithelial lung cancers other than small cell lung cancer and includes squamous cell carcinoma, large cell carcinoma, and adenocarcinoma of the lung.
