As the first step in so many experiments, successful cloning is key to the recovering of meaningful data. In-Fusion PCR Cloning Kit enables directional, seamless cloning of any PCR fragments into any linearized vector in a single 15-minute reaction. No additional treatment of the PCR fragment is needed (no restriction digestion, no ligation, no phosphorylation or blunt-end polishing) for this very efficient kit (over 95%). Furthermore, to simplify the construction and testing of fluorescent protein fusions, Clontech provides prelinearized versions of the popular DsRed-Monomer-N1/C1 and AcGFP1-N1/C1 cloning vectors for fast, efficient PCR cloning with In-Fusion. A single insert can be cloned directly into one of these four In-Fusion Ready vectors, thus saving time and allowing parallel cloning of fusion proteins with different colors at either the N-ter or C-ter.
Stellar Competent Cells lack the gene cluster for cutting foreign methylated DNA, and are therefore useful for cloning methylated DNA. Stellar Competent Cells are ideal for use with Clontech’s In-Fusion Cloning Kits. In-Fusion HD EcoDry Cloning Plus kits are provided in a lyophilized, ready-to-use format that can be stored at room temperature. All the components for the cloning reaction are pre-aliquoted and dried down in tubes. Minimal handling eliminates chances of contamination, pipetting errors, or missing reagents. In-Fusion EcoDry Cloning Plus Systems allow to clone any PCR fragment into any linearized vector in a single step (no restriction digestion of the PCR fragment, no ligation or blunt-end polishing). This ability to clone directly into any vector at any restriction site eliminates the need for further manipulation or subcloning. On the other hand, electrocompetent cells offer high transformation efficiency, making them suitable for many molecular biology applications such as constructing gene banks. The endA1 and recA1 markers in this strain minimize recombination and degradation of plasmid DNA.
Generating libraries for ChIP sequencing (ChIP-seq) can be difficult due to the small amounts of DNA recovered from chromatin immunoprecipitation (ChIP) experiments (particularly against low-abundance transcription factors). The DNA SMART ChIP-Seq Kit adds Illumina-specific sequencing adapters to the ends of DNA fragments in a ligation-independent manner. This DNA SMART ChIP-Seq Kit generates sequencing library for Illumina platforms from 100pg to 10ng of input DNA without using ligation. This highly sensitive kit generates robust sequencing libraries from low-input DNA samples, in just four hours.
